AH2mut mutations disrupt membranous web formation. Ultrathin sections from UHCVcon-57.3 and UHCVcon-AH2mut cells cultured for 48 h in the absence of tetracycline were examined by electron microscopy. While typical membranous webs were found in 17 of 100 sections from UHCVcon-57.3 cells expressing the wild-type (wt) HCV H77 polyprotein, no membranous web was found in 200 sections from UHCVcon-AH2mut cells expressing the H77 polyprotein harboring the AH2mut alanine substitutions in NS4B.
