CamkII activity is required for prehypertrophic and hypertrophic maturation. In situ hybridization analysis of mouse metatarsal cultures treated with inhibitors of CamkII activity (AIP-II and NtideII at the concentrations indicated). Relevant cell types and maturation states were identified as follows: Col9a1 (immature chondrocytes), Fgfr1 (osteoblasts, perichondrium, resting chondrocytes), Prelp1 (proliferative chondrocytes), Ihh (prehypertrophic chondrocytes) and Col10a1 (hypertrophic chondrocytes). The correct size and/or location of these expression domains are indicated by arrows (Fgfr1) or brackets (all others) presented in the solvent control (top panels). AIP-II treatment consistently reduced expression of hypertrophic markers (Ihh and Col110a1) without substantially affecting expression of markers for other maturation zones (Col9a1, Fgfr1 and Prelp1) or non-chondrocyte cell types (Fgfr1). Similar results were observed for NtideII. For all treatments, n=8-12 metatarsals. Scale bar: 200 μm in all panels.
