A) Gating on CD38 and CD138 positive plasma cells (upper left dotplot, gate P2) reveals them to be CD45 negative (upper right dotplot). Selective gating on this CD45 negative plasma cell population (gate P3) reveals them to be monoclonal for cytoplasmic kappa immunoglobulin light chains (lower left dotplot) and to lack expression of lambda cytoplasmic immunoglobulin light chains (lower right dotplot). B) Selective analysis of the kappa light chain restricted plasma cells (gate P4) reveals them to lack expression of CD19 as well as CD45 (lower left dotplot). No lambda light chain positive plasma cells are present in gate P5 as the plasma cells are kappa light chain restricted (lower right dotplot).
