The ERK pathway is required for toxin A–induced IL-8 release. (a) THP-1 cells (106/mL) were preincubated with various concentrations of the MEK inhibitor PD98059 for 30 minutes. Then, cells were stimulated with C. difficile toxin A (100 nM) or LPS (1 μg/mL) for 4 hours. PD98059 partially inhibited IL-8 release induced by toxin A and by LPS (IC50, 2 and 10 μM, respectively). Mean ± SE is shown (n = 3). (b) Peripheral blood monocytes were incubated in the presence of PD98059 for 30 minutes and then stimulated for 4 hours with C. difficile toxin A (2 nM) or LPS (1 μg/mL). PD98059 significantly reduced IL-8 production by both stimuli. Results are expressed as means and SE (n = 3; AP < 0.001; BP < 0.05).
