NKG2D is required for NK cell activation upon adenoviral infection in vivo. (A-B) C57BL/6 mice were treated with 100 μg of anti-NKG2D mAb (+Anti-NKG2D) or the control rat IgG (+Rat IgG) 24 h and 6 h before intravenous injection of Ad-lacZ (Ad). Some mice were left uninfected as controls (Naïve). 24 h after infection, splenocytes were harvested and restimulated with PMA/Inomycin in the presence of Brefeldin A for 4 h. Intracellular IFN-γ and granzyme B was determined by flow cytometry gated on DX5+CD3− NK cells. FACS plots of IFN-γ and granzyme B production with the percentage of IFN-γ and granzyme B positive cells indicated (A).
