Cytoplasmic zinc (Zn) pools were significantly reduced in cells expressing SNP1 but not affected by ectopic expression of SNP2. Cytoplasmic Zn level was measured using a 4X-metal response element (MRE) luciferase reporter construct in cells co-transfected with each SNP and compared with cells transfected to overexpress wild-type ZnT2. Data represent mean luminescence (arbitrary units/μg protein) (n = 3) from 2 independent experiments.
