Expression of Tm-Cyto inhibits the N-CAM-induced NF-κB activation in transfected primary wild-type astrocytes but not in N-CAM-null cells. (A) Immunoblot of astrocytes transfected with pcDNA3.1+ (lane 1), pcDNA-Tm-Cyto (lane 2), or pcDNA-Tm-Cyto/palm (lane 3). Upper bands (140 kDa) are endogenous sd N-CAM; lower bands are the Tm-Cyto proteins. (B) The transfected cells were treated and assayed as described in Materials and Methods. The results are shown as the ratio of the luciferase activity normalized to the β-galactosidase activity for each treatment condition. The error bars represent the standard deviation from quadruplicate samples of a representative experiment. Each experiment was replicated a minimum of four times.
