Jacalin binds O-linked carbohydrate attached to the V1 domain of SIVmac239. (A) SIV04 and Jacalin4 gp120 proteins have markedly lower jacalin binding capacities than that of SIVmac239. HEK293T cells were transfected with no DNA (mock) or with the gp120 eukaryotic expression vector for SIVmac239, SIV04, or Jacalin4. Protein released into serum-free medium was purified with the lectin from Galanthus nivalis. Equivalent amounts of protein from each sample were loaded into six wells of a high-protein-binding plate. Jacalin-HRP was used to probe each sample in quadruplicate. Filled bars represent mean jacalin signals; error bars, standard deviations. Sera from SIV-positive rhesus macaques were used to probe each sample in duplicate. Shaded bars represent signals from sera binding gp120.
