Fig. 1.

Time course of c-jun NH2-terminal kinase (JNK) activation and its relationship to elevation of cytosolic Ca2+ concentration ([Ca2+]i) by the Ca2+ ionophore A-23187 and the Ca2+-ATPase inhibitor thapsigargin. A: effects of A-23187. Rabbit proximal tubular epithelial cells were serum starved overnight and treated with 1 μM A-23187 for the length of time indicated. Cell lysates containing 200 μg protein were immunoprecipitated with anti-JNK1 polyclonal antibody, and the in vitro kinase assay was carried out as described in MATERIALS AND METHODS. Equal protein loading was confirmed by staining the gel with Coomassie brilliant blue before drying and detection of 32P on X-ray film. The autoradiograph is representative of 3 similar results. [Ca2+]i was monitored by fura 2 fluorescence as described in MATERIALS AND METHODS. B: effects of thapsigargin. Cells were treated and analyzed as in A, except that 1 μM thapsigargin replaced A-23187. pATF-2; phosphorylated ATF-2 detected by 37P incorporation.
