Correlation of Ca2+-chelating affinity with JNK activation potency. A: structure and Ca2+ dissociation constants for BAPTA and dimethyl BAPTA. B: time course of JNK activation by BAPTA-AM and dimethyl BAPTA-AM. Quiescent cells were treated with 10 μM BAPTA-AM or dimethyl BAPTA-AM for the times indicated. JNK activity was measured by the in vitro kinase assay. C: dose-response data for JNK activation by BAPTA-AM and dimethyl BAPTA-AM at 30 min. JNK activity was measured by immunoblot with anti-phospho JNK antibody. Results from 1 series of experiments (out of 2) are shown; the results of the 2 series were similar.
