Effect of inhibition of Ca2+ influx on BAPTA-AM- and thapsigargin-dependent activation of JNK. A: effect of SKF-96365 and nifedipine. Cells were serum starved overnight and pretreated with 10 μM SKF-96365 or 10 μM nifedipine for 20 min, before exposure to 10 μM BAPTA-AM for 30 min or 1 μM thapsigargin for 15 min. JNK activity was measured by the in vitro kinase assay. Data are presented as percentage of unstimulated JNK activity (=100%). Significance was determined between stimulation in the presence or absence of each inhibitor. *P < 0.05. B: effect of removal of free, extracellular Ca2+ with EGTA. Cells were stimulated with 10 μM BAPTA-AM for 30 min or 1 μM thapsigargin for 15 min in the presence or absence of 3 mM EGTA in the medium. Data shown is a representative of in vitro kinase assay.
