Cleavage of C3b to iC3b occurs in the presence of fH and fI. S. aureus were incubated with 10% heat-inactivated serum for 30 mins at 37°C to bind serum fH. After washing, various combinations of fI, C3b or C3 (1 ug each) were added and allowed to incubate for 1 hr at 37°C. Controls included bacteria not incubated with serum, and samples of purified fH and fI with purified C3 or C3b. The bacteria were sedimented and the supernatant was analyzed for the presence of C3b and iC3b via Western blotting, under reducing conditions; the types of C3 chains are indicated (α’, β, α’2).
