Thirty–50 mg of parotid gland tissue from AAV5 (herein) and AAV28-vector transduced macaques were suspended in Cell Culture Lysis Reagent (Promega) and homogenized with an Omni TH 1 homogenizer to extract protein. Fifty μl of this extract were used to determine the presence of RhEpo protein with a human erythropoietin ELISA kit (StemCell Technologies, Vancouver, BC, Canada). The concentration of protein was determined using the BCA protein assay (Pierce, Rockford, Il, USA). Each gland sample, indicated by animal number and the vector administered, was tested in duplicate and average values are shown.
