 Knockdown of p190A RhoGAP restores stress fibers and FAs in Gab2-overexpressing cells. (A) Reduction in p190A RhoGAP protein levels following transfection of targeting siRNAs. MCF-10A/Gab2 cells were transfected with the indicated siRNAs (see Materials and Methods), alone or in combination, and cell lysates were then Western blotted as indicated. NT indicates nontargeting control siRNA. (B) Knockdown of p190A RhoGAP reverses the Gab2-associated cellular phenotype. Cells were transfected as indicated, plated on collagen IV for 18 h, and then stained for actin with phalloidin or with anti-paxillin antibodies. (C) Scoring of FA size and distribution. Approximately 800 cells were classified from two independent experiments. Data represent the mean. For classical FAs, variation from the mean was <10% for vector cells and <4% for the other transfections.
