Knockdown of REST/CoREST protein levels. U251 cells in 25-cm² flasks were either mock transfected or transfected with 200 pmol of REST or CoREST siRNA or irrelevant (control) siRNA. The cells were harvested at 48 h after transfection. Lysates containing 70 µg of total proteins were separated on denaturing 8% polyacrylamide gels, electrically transferred to nitrocellulose sheets, and immunoblotted for REST, CoREST, LSD1, Sin3A, and β-actin proteins, as described in Materials and Methods. The images were scanned with the aid of ImageJ, and the density of the bands was normalized with respect to the density of the proteins in mock-treated infected cells.
