(A) Western Blot analysis demonstrating downregulation of ER-α. BCPAP and MCF-7 cells were transfected with siRNA and whole cell protein was resolved by SDS-PAGE followed by Western blot analysis for ER-α. A decrease in ER-α expression was observed for both BCPAP and MCF-7 confirming ER-α silencing. Actin was used as a loading control. (B & C) Cancer cells were transfected with ER-α siRNA and subsequently treated with 10−8 M E2±10−6 M fulvestrant ±25 µM DIM or left untreated. Data presented demonstrates percent migration (B) and invasion (C) of ER-α silenced cancer cells treated with E2± fulvestrant ± DIM compared to untreated ER-α silenced cancer cells which were set to 100%. MCF-7 was used as positive control for ER silencing experiments.
