The dga1Δlro1Δare1Δare2Δ quadruple mutant exhibits inositol auxotrophy at 37 °C. Overnight cultures grown in I⁺ or I⁻ medium at 30 °C were diluted back to A₆₀₀ = 0. 15 in 50 ml of the same medium and allowed to grow to mid-logarithmic phase at 30 °C. Samples were harvested and resuspended at a concentration of 0.5 A₆₀₀. Each sample was diluted in multi-well plates by 1:10 serial dilutions, and 10 μl of cells from each dilution were spotted on I⁺C⁻, I⁻C⁻, I⁺C⁺, and I⁻C⁺ plates and allowed to grow at the designated temperatures for 3 days.
