Chromatin condenses rapidly during thymocyte development. (A) Thymocytes were isolated and stained with DAPI and with antibodies to mono-, di- and tri-methylated histone H3K4, tri-methylated histone H3K9/27 and acetylated histone H3 to detect modification of histone H3. Antibodies to N-terminal and C-terminal epitopes of unmodified histone H3 are also shown. (B) Flow cytometric analysis of chromatin condensation during thymocyte development using PE- conjugated H3K4me1 antibodies as a marker for chromatin accessibility. Subpopulations of Thy1.2+ thymocytes were delineated based on surface expression of CD4 and CD8.
