Outline of the tandem-affinity-immunopurification strategy. An artificial adaptor protein combines an IGB with a calmodulin-binding peptide (C) and a strep tag (S). The adaptor is used to immobilize antibodies on calmodulin beads, which are then incubated with extracts containing the antigen. Immunocomplexes are recovered by native elution with EGTA and are re-purified on Streptactin beads.
