After an overnight incubation in the retrovirus-containing medium, Col11a2-Egfp-Ires-Puro transgenic MDFs were replated into the 6-well plates (day 1) and subjected to time-lapse observation by Biostation CT (Nikon). Phase and GFP images were taken once a day. On day 1, MDFs did not express GFP. On day 8, nodules (arrows) appeared and expressed GFP. The magnification of the boxed region in the phase image on day 8 (upper middle panel) showed that the nodules formed multiple layers of cells and consisted of polygon-shaped cells (upper right panel).
