Transformants were grown in SC-leu medium to exponential phase and run on transcription assays were performed as described in Materials and Methods. Radioactivity incorporated into the G-less cassettes was quantified in a Fuji FLA-5100. For each sample, the ratio of the total numbers of counts incorporated into the distal versus the proximal G-less cassette was determined and normalized against the ratio for the same construct in the wild-type strain. The average and SD of three independent experiments are shown. The values to the left are molecular sizes in nucleotides.
