Reaction of the anti-glutamate MAb 215B2 with amino acids fixed in situ with 4% paraformaldehyde or with 4% paraformaldehyde/0.3% glutaraldehyde in wells of ELISA plates coated with rat brain tissue lysate. Glutamate (black bar) is fixed in situ by the mixed aldehyde solution, but not by paraformaldehyde alone, and the fixed glutamate is specifically recognized by the anti-glutamate monoclonal antibody MAb 215B2. The error bars represent standard deviations of triplicate assays; MAb 215B2 binding in wells receiving glutamate differed from all controls (p<0.025, t-test).
