FIGURE 6. — Confocal microscopy analysis of arrestin distribution upon agonist activation. HeLa cells co-transfected with FLAG-β₂AR and HA-arrestin-3-WT (A) or HA-Arrestin-3–4K/R (B) were serum-starved before being treated with vehicle (0.01 mm ascorbic acid) and 10 μm isoproterenol (ISO) for 5 min. The cells were then fixed, permeabilized, and immunostained with anti-FLAG and anti-HA antibodies. Yellow in the merged images represent co-localization between FLAG-β₂AR and HA-arrestin-3. The differential interference contrast (DIC) image is shown. Shown are representative images from cells analyzed from three independent experiments in which at least 10 cells were analyzed per experiment. Bar, 20 μm.
