Lucifer yellow uptake and FM4-64 uptake are increased in hgt1-Δ cells transformed with the five human genes. After being cultured at 30°C in minimal selective medium to a cell density (expressed as OD₆₀₀) of 0.5, cells were washed and grown for another 11–12 h at 30°C in sulfur-deficient medium in the absence of GSH. To start the uptake experiment, cells were incubated with 4 mg/ml lucifer yellow or 40 μM FM4-64 at 30°C for 2 h, shaking at a speed of 200 rpm for Lucifer yellow or 75 rpm for FM4-64.
