PY motif mutations largely abolish the effect of LITAF on intracellular GSH levels, and on [3H]GSH and FM4-64 uptake. PPSY motifs in LITAF were changed to PPSA by site-directed mutagenesis. Wild type strain ABC154 was transformed with empty vector, LITAF, or LITAF with PY mutations and whole cell lysate of each transformant was subjected to SDS-PAGE gel. The blot was probed with anti-FLAG antibody for LITAF expression, and with anti-actin for loading control (9A). Values are given as means ± SEM, N=3-6. * Significantly different intracellular glutathione concentration (9B), [3H]GSH uptake (9C) and FM4-64 uptake (9D) when compared to wild type cells transformed with the empty pESC-URA vector (p<0.05).
