PY motif mutations largely abolish the effect of LITAF on intracellular GSH levels, and on [³H]GSH and FM4-64 uptake. PPSY motifs in LITAF were changed to PPSA by site-directed mutagenesis. Wild type strain ABC154 was transformed with empty vector, LITAF, or LITAF with PY mutations and whole cell lysate of each transformant was subjected to SDS-PAGE gel. The blot was probed with anti-FLAG antibody for LITAF expression, and with anti-actin for loading control (9A).
