Cumulative dose-effect curves for pregnanolone (A), flunitrazepam (B), midazolam (C), pentobarbital (D), ketamine (E), and morphine (F) before (circles), during (triangles: pregnanolone last administered 1 hour prior to session; squares: pregnanolone last administered 25 hours prior to session), and after (diamonds) chronic pregnanolone treatment. During chronic treatment, dose-effect curves obtained when pregnanolone was last given 1 hr prior to the session (triangles) were determined during week 11 for pregnanolone (A) and flunitrazepam (B) and during week 12 for midazolam (C) and ketamine (E), before tolerance developed to the rate-increasing effects of pregnanolone (points above V); dose-effect curves for pentobarbital (D) and morphine (F) were determined during week 16 of treatment after tolerance developed to the rate-increasing effects of pregnanolone.
