Table 1. Kinetics of the parasites' death under H2O2 treatment.

  AnV (+) 7-AAD (−) AnV (+) 7-AAD (+) AnV (−) 7-AAD (+)
Wild-type (−) H2O2 0.033 0.007 0.004
Wild-type (+) H2O2 0.14 0.42 0.79
CPC KO (−) H2O2 0.29 0.07 0.02
CPC KO (+) H2O2 0.29 0.28 0.18
CPC KO/+ (−) H2O2 0.16 0.04 0.02
CPC KO/+ (+) H2O2 0.32 0.45 0.53

Wild-type parasites (WT), cathepsin B-deficient parasites (cpc KO) and cpc KO complemented strain (cpc KO/+) that contain an episomal copy of cpc were not treated (−) or treated (+) with 1 mM H2O2 for 1 h. Cells were stained with fluorescein isothiocyanate (FITC)-labeled Annexin V and 7-AAD to distinguish between live (AnV−/7AAD−), apoptotic (AnV+/7AAD−, early apoptotic cells; AnV+/7AAD+, late apoptotic/necrotic) and necrotic cells (AnV−/7AAD+) as depicted in Figure 6. Values correspond to ratios of percentage of AnV+/7-AAD−, AnV+/7-AAD+ and AnV−/7-AAD+ cells to percentage of AnV−/7-AAD−
