TOR controls the localization of the kinases TPK1 and YAK1. (A) Wild-type (wt, TB50a), bcy1 (TS141), and sit4 (TS65-2D) cells transformed with pTS134 (HA2-TPK1) or wild-type cells transformed with pTS137 (HA-BCY1) were pregrown in selective synthetic medium, shifted to YPD, grown for three or four generations, and treated with drug vehicle or rapamycin for 45 min. They were then subjected to indirect immunofluorescence analysis to visualize TPK1 or BCY1, respectively. The exposure time used to visualize TPK1 in bcy1 cells was approximately fourfold longer than for wild-type cells. The cellular distribution of HA2-TPK1 or HA-BCY1, respectively, is shown in the left column; the same field of cells is shown in the middle (Nomarski) and right (DNA; DAPI staining) columns. (B) Wild-type cells expressing YAK1-myc13 (TS129-8C) were grown to logarithmic phase in YPD medium and treated with drug vehicle or rapamycin for 30 min.
