hMADS cells were synchronised as described in “Methods” and treated with 5 µM cyclopamine. After 24 hours cyclopamine was removed and the subconfluent cells were treated with an adipocyte or an osteoblast differentiation cocktail. After 10 days mRNA were extracted and aP2, PPARγ, SREBP1-c, Runx-2, osteoprotegerin and ALPL expressions were evaluated by quantitative RT-PCR. Results are representative of three independent experiments.
