Yeast growth assay used to establish optimal conditions for the planned MbYTH screen. The pAMBV4-M3-Cub plasmid was transformed into the NMY51 yeast strain, together with the indicated control plasmids [Alg5-NubI, Alg5-NubG, or pADL-Nx (empty vector)]. Yeast growth was measured by determining the absorbance at 630 nm. The compositions of the media used are indicated in the inset. Yeast growth was determined in the absence or presence of the indicated concentrations of 3-AT, an agent that blocks one of the key enzymes involved in histidine biosynthesis. The data shown are representative of three independent experiments, each carried out in triplicate.
