Effect of siRNA-mediated knock-down of Tmem147 expression on the M3R-mediated increase in H508 cell proliferation. Cells were treated with negative control siRNA or Tmem147 siRNA, as described in detail under Materials and Methods. Cells were grown for 5 days in the absence or presence of the muscarinic agonist carbachol (100 μM) either in the absence or presence of the muscarinic antagonist atropine (1 μM). Cell growth was determined by using a colorimetric assay (see Materials and Methods for details). In each individual experiment, cell growth observed with cells that had been treated with control siRNA and had not been exposed to carbachol was set equal to 100%. Results are expressed as means ± S.E.M. of four separate experiments (*, p < 0.05).
