The full-length HA and NP genes of H5N1-HPAIV strain A/Chicken/Henan/12/2004 was cloned into the dual-promoter plasmid pBudCE4.1 for eukaryotic expression. (B) Expression of DNA constructs in HEK293T cells. The HEK293T cells were transfected with equal molar amounts of the indicated plasmids, and cell lysates were collected 24 h post-transfection and subjected to Western blot analysis with anti-HA and anti-NP polyclonal antibodies. The β-actin was detected with anti-actin-specific polyclonal antibody as a loading control.
