Disruption of both IN-TNPO3 and of IN-importin α complexes by the Rev protein. Rev-expressing SupT1 lymphocytes were infected with wild-type HIV-1 and then half the volume of cell lysate was subjected to SDS-PAGE and immunoblotting with either anti-importin α (anti-Impα), anti-TNPO3, anti-IN or anti-Rev antibodies. Complementary HRP-conjugated antibodies were used as second antibodies. The remaining lysate was subjected to co-IP with anti-Impα, anti-TNPO3, anti-IN or anti-Rev antibodies and the complementary HRP-conjugated antibodies as second antibodies.
