CCR5 and CD4 expression levels at the surface of cells infected by the JRRen or the Bx08Ren viruses and IC50 values for MVC or TAK779 in infection inhibition assays

The IC50 values for TAK779- and MVC-mediated inhibition of infection were calculated by nonlinear regression analysis of the experimental data presented in Fig. 6. Cell surface expression levels of CCR5 and CD4 (geometric mean fluorescence intensities) were measured by flow cytometry analysis using the phycoerythrin-conjugated mAbs 2D7 and SK3 (BD Biosciences), respectively. Background staining was assessed with the appropriate isotype- and fluorochrome-matched control mAb and subtracted. Analysis was performed on a FACSCalibur flow cytometer (BD Biosciences). Results represent means ± S.D. of four independent experiments.
