Enhanced tumor responsiveness of DGKζ−/− CD8+ T cells. 2 × 105 naive (CD44low), CD45.1− OT-I, or DGKζ−/− OT-I cells were adoptively transferred into CD45.1-positive mice. Forty eight hours later, 5 × 105 EL4-ova cells were injected subcutaneously into the right flanks of host mice. Seven days later, mice were sacrificed, and tumors were excised and weighed (top panel). Splenocytes were prepared and donor lymphocytes were assessed for the up-regulation of CD44 on donor CD8+ T cells (middle panel), or stimulated for 5 h with OVAp and assessed for the production of IL2 in donor cells as described for Fig. 4 (bottom panel).
