Peptide NMQALSMPVT blocks binding of heparan sulfate to mNdst1. A, emission spectra were obtained for 20 μm mNdst1 mixed with 50 μm heparan sulfate (□), 400 μm NMQALSMPVT (♦), 400 μm NMQALSMPVT, and 50 μm heparan sulfate (▿) or 400 μm NMQALSMPVT and 200 μm heparan sulfate (■). B, mNdst1 (2 μg) produced in wild type cells radiolabeled with [35S]sulfate was mixed with the indicated concentrations of the peptides. The beads were sedimented by centrifugation, washed, and analyzed by agarose gel electrophoresis. Peptide NMQALSMPVT displaced bound [35S]heparan sulfate above 50 μm, whereas peptide RGWRGEKIGN was only partially effective.
