Limited SmaI digestion of URF (A) or SRF (B) chromosome blocks. Blocks were digested at 25°C overnight with varying units of SmaI: lane 1, 0 U; lane 2, 0.01 U; lane 3, 0.1 U; lane 4, 1 U; lane 5, 2 U; lane 6, 3 U, and subjected to an 18 h PFGE procedure as described in the Materials and Methods.
