CD55, but not Thy-1, is expressed on human peripheral blood invariant natural killer (iNKT) cells. Peripheral blood was collected from healthy donors and mononuclear cells were isolated using a Ficoll-Paque gradient. Cells were stained with a FITC-conjugated anti-CD3 monoclonal antibody (mAb), allophycocyanin-conjugated human PBS-57-unloaded (control) or -loaded CD1d tetramer, and a phycoerythrin-conjugated anti-Thy-1 mAb (open histogram) or isotype control (filled histogram). CD3⁺ CD1d tetramer⁺iNKT cells were gated on and Thy-1 expression was analysed by flow cytometry (a).
