FIGURE 2.

The role of GluR6-containing kainate receptors in receptor-coupled Bcl-2 denitrosylation. A, effects of the GluR6 inhibitor NS102 upon KA-induced Bcl-2 denitrosylation at 3 h post-treatment. The samples were processed using the biotin switch method followed by Western blotting. B, effects of GSNO, SNP, and DTT on Bcl-2 S-nitrosylation in KA-treated (3 h) CA1 and CA3/DG subregions of the rat hippocampus. GSNO and SNP are NO donors; DTT is an NO inhibitor. The samples were processed using the biotin switch method followed by Western blotting. C, effects of KA in the presence or absence of NO modulator SNP, GSNO, DTT, and antioxidant NAC on Bcl-2 phosphorylation at 3 h post-treatment. The bands were scanned, and the intensities were determined by optical density measurements. The data are the means ± S.D. of the fold changes with respect to the saline control. *, p < 0.05 versus the saline group; #, p < 0.05 versus the respective DTT pretreated group (n = 3).
