A weak cytoplasmic dsRNA response is a general feature of pluripotency. (A) Differentiated hESCs gain an IFNβ response to PIC. H9 cells and BMP4-treated H9 cells were transfected with 2 µg/mL PIC for 6 hrs or 24 hrs. IFNβ responses were quantitatively measured by RT-QPCR and normalized to each endogenous actin mRNA. Experiments were at least duplicated and error bars were calculated from three biological repeats. (B) Reprogrammed iPS cells lack an IFNβ response to PIC. iPS (IMR90) cells were treated with either 50 µg/mL PIC directly, or transfected with 2 µg/mL PIC for 6 hrs or 24 hrs. The IFNβ responses were quantitatively measured by RT-QPCR and normalized to each endogenous actin mRNA.
