Structural organization of MDGA1 and truncated proteins. MDGA1 full length (pFLAG-MDGA1) contains an N-terminal signal peptide followed by flag sequence, six immunoglobulin-like (Ig) domains, one single fibronectin type III (FnIII) domain, a MAM (Meprin, A5 protein, receptor protein-tyrosine phosphatase μ) domain and a potential cleavage site (Asp⁹³¹) for GPI (GlycosylPhosphatidylInositol) in the C-terminal anchoring the protein to the cell membrane. In the truncated proteins we have removed either the MAM domain (pFLAG-IgGPI) or the six Inmunoglobulin and the FnIII domains (pFLAG-MAMGPI)
