VacA targets mitochondria. (a) Wt MEF cells were incubated for 24 h with wt or P9A mutant H. pylori supernatants, harvested and subfractionated as described. Fifteen or 50 μg of early endosomal (EE), late endosomal (LE) and mitochondrial (Mt) proteins were separated by SDS-PAGE and immunoblotted using the indicated antibody. (b) Wt MEF cells, transfected with plasmid encoding for mtRFP, were incubated with wt or P9A mutant H. pylori CS. After 24 h of incubation, cells were fixed and the toxin was stained with a rabbit anti-VacA antibody followed by a secondary FITC anti-rabbit antibody.
