Transient transfection with a dominant-negative Src attenuates EMCV-induced COX-2 expression by macrophages. RAW264.7 cells (2×106) were electroporated with 2 μg dominant-negative Src plasmid (dnSrc) or a vector control, or were mock transfected. Transfected cells (4×105 in 400 μl DMEM per condition) were treated with EMCV (m.o.i. 1) for 24 h. Total cell lysates were harvested and protein expression was determined by Western blot analysis. The results are representative of three independent experiments. GAPDH was used as a loading control.
