Cld4 shows negligible homotypic cell adhesion activity. (A) Cultured Eph4 epithelial cell lines and freshly isolated newborn thymocytes were fixed and two-color stained with the indicated antibodies. (B) Cld thymocyte cell lines (SCID.adh) transfected with Cldn1 or Cldn4 were stained with the corresponding and anti-CD45 antibodies. Phase-contrast images are also shown. Arrows indicate the accumulation of Cld1 at the cell contact sites. (C) The Pt/C replicas of SCID.adh cells transduced with Cldn4 or control vector (GFP) were stained with anti-Cld4 antibody followed by colloidal gold-conjugated protein A and examined with EM.
