Knockdown of Cldn4 results in the reduced generation of SP cells in FTOC. The E15 thymocytes were infected with pSIREN retrovirus containing Cldn4 shRNA (pSIREN-Cld4) or scrambled oligonucleotides (pSIREN-cont) and cultured with the dGuo-treated E15 thymic lobes.
