Quantification of the expression of GFAP and TNFα in the vlPAG in the morphine withdrawal. The expression of GFAP and TNFα in the vlPAG was evaluated by calculating the relative density of GFAP immunostaining with the NIS-Elements Basic Research software (Nikon). In sham group (T0Z+S), animals received microinjection of T0Z into PAG and chronic saline (1 ml, twice a day, i.p.) for 5 days; 1 h after last injection of saline, animals received naloxone (4 mg/kg, i.p.). The percentage of density from the sham group was calculated as 100%. T0TNFSR (TNFSR+M), but not T0Z (T0Z+M) injection, significantly suppressed the activation of GFAP in the PAG, **p<0.01 vs sham, ##p<0.01 vs T0Z, n=4–6, ANOVA. T0TNFSR injection significantly suppressed the activation of TNFα in the PAG, **p<0.01 vs sham, ##p<0.01 vs T0Z, n=4–6, ANOVA. Scale bar, 100 μm.
