Table 1. In Vitro Estimates of D2/D3 Selectivity of Antipsychotics and the Predicted Occupancy at D3 Receptors at Clinically Effective Levels of D2 Receptor Blockade Based on Previous Reports (Levant, 1997; Schotte et al, 1996).
| Antipsychotic | No. of studies | D2 : D3 Selectivity±SD | D2 Occupancy | Predicted D3 occupancy |
|---|---|---|---|---|
| Clozapine | 9 | 2.82±2.01 | 60% | 35% |
| Haloperidol | 11 | 3.03±4.42 | 75% | 50% |
| Risperidone | 3 | 4.9±3.71 | 75% | 38% |
| Olanzapine | 1 | 3.18 | 75% | 49% |
| Amisulpride 230 mg | 3 | 2.29±0.58 | 67% | 47% |
| Amisulpride 670 mg | 3 | 2.29±0.58 | 80% | 64% |
| Raclopride | 3 | 1.16±0.53 | — | — |
D2, and predicted D3, occupancies are for illustrative purposes and are intended to reflect the normal therapeutic range for each compound. The calculation suggests that D3 occupancy should range from 1/2 to 2/3 of D2 occupancy at these levels of D2 occupancy.
