Cells expressing both GFP-Sro7 (CEN) and RFP-Sec2 (CEN) were grown in selective media overnight and then fixed and processed for fluorescence microscopy. The merged image of GFP-Sro7 and RFP-Sec2 fluorescence is shown on the right. Bar, 1 μm. (D) Wild-type, sec6-4, sec2-41, and myo2-66 mutant strains containing GFP-Sro7 were grown in selective media to midlog at 25°C and then shifted to 37°C for 1 h before fixing and processing for fluorescence microscopy. Quantitation of GFP-Sro7 polarization was obtained by scoring a minimum of 50 cells for the presence of GFP-Sro7 at emerging bud sites and bud tips.
