In vitro binding assays were conducted with GDP- or GTPγS-loaded Sec4 immobilized on glutathione Sepharose beads, soluble Sro7 (1 μM) and soluble wild-type IQ region of Myo2 (4 μM), or mutant Myo2IQ1,5 (4 μM). Graph shows results from four independent experiments, and SD and a Student’s t test showed a p value < 0.016. (B) Model for the proposed function of Myo2 in Sro7 regulation of post-Golgi vesicle docking and fusion or in vesicle clustering.
