Lucanthone induces cathepsin D expression and decreases cell viability independently of p53. A, HCT116 p53+/+ and p53−/− cells were used to evaluate whether p53 was required for lucanthone-mediated apoptosis. Immunoblotting confirmed the lack of p53 in the HCT116 p53−/− cell line. 72 h MTT assay determined that HCT116 p53−/− cells were less sensitive to etoposide. Mean ± S.D., n = 3. * indicates a significant difference compared with HCT116 p53+/+-treated cells. B, lucanthone increases cathepsin D levels in HCT116 cells independent of p53 status. Cells were treated with 5 μm lucanthone for 48 h. Cathepsin D was visualized by immunocytochemistry and quantified using ImageJ software. Mean ± S.D., n = 5.
